Mines a 1x10 setup, I really like it, saves weight and make everything look so much cleaner! It is more difficult to climb on steep hills but you DO get stronger and it becomes easier. The range of gears is perfect. I would totally recomend it! As for the wheels... The only difference I've felt is that the niner rolls better that 650b. The 29er I had b4 was a xc, so the tires were not as wide. Oh and 650b is ALOT more nimble than 9er. (Do you have a mtb? And what kind?)
