Where the rim goes up you need to tighten the spokes evenly, where it dips down you need to slacken the spokes. If you have a dip but no rise on the opposite side of the wheel you have a flat spot which you'll struggle to get out.
The easiest way to do it is to de tension all of your spokes by a turn or 2 (gradually, start at the valve, quarter turn on each spoke). Then concentrate on the radial true. Don't worry too much about lateral. Once you get rid of the up and down, then true it laterally. Then dish it last
